A lot that tests at 99% purity on arrival can be materially degraded a fortnight later if it is handled carelessly. Stability is a property of the whole chain — transit, storage, reconstitution, and the container the material ends up in — not just of the synthesis. This note covers laboratory handling of lyophilised research compounds.
Scope: this is a materials-handling note for in-vitro laboratory work. It describes preparing and storing compounds for bench use. It is not guidance for any use in humans or animals, which is prohibited under our Terms of Service.
On arrival
- Inspect the outer packaging and the vials before anything else. Photograph any damage before opening — most replacement policies, ours included, run on a short reporting window.
- Check that the lyophilised cake is intact. A cake that has collapsed into a film or a visible residue on the stopper suggests the vial warmed in transit.
- Confirm the lot number on the vial matches the certificate published for that batch.
- Move vials to storage promptly rather than leaving them on a bench.
Storing lyophilised material
In the freeze-dried state most peptides are comparatively robust; the enemies are moisture, heat, light and repeated temperature cycling.
- Cold and dark. Follow the storage condition stated on the certificate for that compound. Keep vials in their original container, away from light.
- Dry. Lyophilised powder is hygroscopic. Let a vial reach room temperature before opening, or condensation will form inside it the moment the seal is broken.
- Stable, not cycled. Repeated warming and cooling is more damaging than a slightly higher constant temperature. Take out what you need rather than moving the whole stock.
- Labelled. Keep the lot number with the vial. Once it is separated from its packaging, its certificate no longer means anything.
Reconstitution technique
Reconstitution is where most avoidable losses happen. Peptides are surface-active and shear-sensitive; the way liquid is introduced matters as much as what is introduced.
- Bring the vial fully to room temperature before opening.
- Wipe the stopper with an alcohol swab and let it dry.
- Introduce the solvent slowly, directed down the inner wall of the vial rather than jetted onto the cake.
- Do not shake. Swirl gently, or leave the vial to stand until the cake dissolves. Shaking denatures peptides and generates foam, and material trapped in foam is material lost.
- Inspect the solution. It should be clear. Cloudiness or visible particulate means incomplete dissolution or precipitation.
- Record the date, the solvent and the concentration on the vial.
Bacteriostatic water containing 0.9% benzyl alcohol is the common choice for multi-draw work because the preservative suppresses microbial growth in an opened vial. Sterile water suits single-use preparations. Some compounds require a different solvent entirely — check the certificate and the product page before assuming water is appropriate. After reconstitution
- Solutions are considerably less stable than powder. Plan work around a short in-solution window rather than a long one.
- Refrigerate; do not leave solutions at room temperature between sessions.
- Avoid freeze–thaw cycling. If material must be frozen, aliquot first so each aliquot is thawed once.
- Use low-binding tubes where concentration is low. At dilute concentrations, adsorption onto ordinary plastic is a real source of loss.
- Keep the lot number attached to every aliquot, so any result can be traced back to a certificate.
Why this belongs in your records
If an experiment produces an unexpected result, the first question is whether the material was what you thought it was. That question is answerable only if the lot number, the certificate, the reconstitution date, the solvent and the storage history are all written down. A certificate establishes the starting condition; your own records establish everything after it.
